Wound Healing by Allogeneic Transplantation of Specific Subpopulation From Human Umbilical Cord Mesenchymal Stem Cells

In normal physiological conditions, restoration of a functional epidermal barrier is highly efficient; nevertheless, when it fails, one the main consequences chronic ulcerative skin defect, most frequently recognized complications diabetes. Most these venous ulcers do not heal with conventional treatment, leading to appearance infections and in patient. Treatments based on use autologous mesenchymal stem cells (MSC) have been successful; however, its implementation entails complications. The umbilical cord offers an unlimited source adult MSC (ucMSC) from Wharton’s jelly tissue same relevant features for clinical applicability avoiding difficulties. It has recently characterized by specific subpopulation derived ucMSC, differentiated (DMCs). This expresses human leukocyte antigen-G (HLA-G) molecule, strong immunosuppressive checkpoint, vascular endothelial growth factor (VEGF), potent angiogenic factor. Considering importance developing more effective therapy wound especially lesions, we analyzed DMC safety, efficacy, therapeutic potential. By immunohistochemistry, cords HLA-G VEGF positive were selected. Flow cytometry revealed that 90% are HLA-G+, CD44+, CD73+, CD29+, CD105+, CD90+, HLA-DR−. Reverse transcription-polymerase chain reaction expression all subpopulations.
